Accelerating GMP Readiness Through Risk-Based CQV and Strategic Execution
Brevitas deployed a risk-based CQV strategy leveraging GxP test prioritization, clear system boundaries, leveraging commissioning data, proactive QRM, and cross-functional execution—qualifying 20+ cleanrooms and validating the EMS under compressed timelines.
Business Challenge
Our client’s GMP facility faced construction delays and a growing list of unresolved punch list items, jeopardizing commissioning timelines and cleanroom release for EMPQ. Simultaneously, the Environmental Monitoring System (EMS) was in qualification, and critical maintenance and operational procedures were not yet in place, raising compliance and readiness risks.
Our Approach
To recover the timeline and ensure GMP readiness, Brevitas implemented a proactive, risk-based CQV strategy designed to accelerate cleanroom qualification while maintaining full compliance. The team emphasized risk management, cross-functional alignment, and strategic reuse of available data to reduce rework and resource strain.
Key elements of the approach included:
- Streamlined Qualification Boundaries
Clearly defined system boundaries enabled the reuse of commissioning data for Installation Qualification (IQ), reducing redundancy. Review of EMS validation (e.g., RH, temperature, and differential pressure probes) eliminating unnecessary retesting and documentation effort.
- GxP-Driven Prioritization
Tasks were categorized as GxP vs. non-GxP, allowing efficient scheduling and resource allocation. This reduced training burden, minimized rework, and enabled focused execution on compliance-critical systems.
- Proactive Quality Risk Management (QRM) & Contingency Planning
Early identification of decision points, failure scenarios, and alternate workflows helped mitigate non-conformances and avoid timeline bottlenecks. This created flexibility in execution while upholding quality standards.
- Integrated Cross-Functional Collaboration
Brevitas coordinated closely with contractors, engineering, quality, and operations teams to resolve punch list items, manage change controls, and maintain traceability across all qualification activities.
- Dry Runs & Extended Support Windows
Protocol dry runs were executed to catch documentation issues and overlooked punch items before formal execution. Weekend and off-shift support provided the client with extended availability, helping reduce idle time and accelerate milestone completion.
- Controlled Cleanroom Release Based on EMPQ Readiness
A phased approach to room qualification was adopted, prioritizing cleanrooms and supporting systems required for Environmental Monitoring Performance Qualification (EMPQ). This enabled timely readiness for microbial sampling and reduced critical path dependencies.
This structured and responsive approach delivered measurable time and cost savings, reduced validation-related errors, and enhanced compliance outcomes—ensuring the facility progressed toward GMP readiness with minimal disruption.
Results Achieved
- Timeline Recovery & Acceleration: A 1-month compression in project timeline through risk-based cleanroom release, concurrent EMS and room qualifications, and decisive change management.
- Successful Qualification of 20+ Cleanrooms: Including ISO 7 and ISO 8 areas, enabling on-time EMPQ initiation and alignment with target go-live.
- Minimized Rework and Deviations: Strategic reuse of commissioning data, clear boundary setting, and protocol dry runs led to fewer punch list escalations.
- Enhanced Cross-Functional Readiness: Aligned contractors, QA, engineering, and operations into unified execution model, reducing onboarding, documentation errors, and post-execution clarifications.
